during the first fight with Yselda, we need to keep the Megasylvan Sapling (all of them) alive in order to complete the quest and get the flower(violet tulip). Should we do this every time, when we fighting with boss? I wonder if we'll be kick out of the room when the plants die. I don't mean the fight to end quest, just like make daily boss.

Based on Tibia Fandom Quest update:

There are two possible ends to this fight. If you simply kill Yselda and let any of her Megasylvan Saplings die (there's four in total, one in each corner), you'll still be able to complete part of the quest. To go even further, however, you need to keep her four Megasylvan Saplings alive the entire fight.

There is a change in the dialogue with Kesar also and you will not receive the Tulip.

so... I mean just the daily boss fight. does that mean I don't have to keep plants in combat when I just want loot?

Also once completed fight Yselda one more time and after killing her use tulip next to her for achievement.
